My starter kit came with 2 buckets, one for fermenting and one for bottling. Both have a spigot/tap. Is it ok to take a gravity sample from the spigot on the fermenter? Will the trub that I'll probably get in the sample affect the gravity reading?

It's OK to take a sample from that. As long as the trub settles to the bottom of your hydrometer cylinder, and below where the hydrometer settles, it won't affect the reading.
