Quick question, so i propagated up dregs successfully from a 16oz can about a month ago. They are stored in a mason jar in my fridge. I am unsure of the cell count so i thought i would make a 2 step starter for to use in my next batch and pitch half then save the other half. Is that the best practice? I have heard you can weigh out the yeast slurry and get an estimate of the cell count that way. Not sure if that is the best way or not?

What does everyone else do to get in the ballpark with the correct pitch rate?
 
I have done pretty much what you describe - step it up a couple times, after which I figure I have a pretty good idea of what the cell count is.
