So for my birthday my wife bought me a 3 gallon pin lock keg and gas line and beer line connections and tubing. I have one tap right now and my co2 tank only has a single regulator on it. What it the best way to set this up

Get a manifold. Just get a 4-port manifold, you'll need them later :)

With regards to your taps, not sure what to tell you there. If you have a tower, you could invest in a dual head tower. If you don't want to deal with conversion atm, just get a picnic tap and coil it on top of the keg. picnic taps are kinda messy, but they work till you improve the delivery system.
 
Will I be able to have different serving pressures with that. As well as taking on and off the gas will it effect the beer if I get a party tap and have a single tank and manifold.

Nope. If you want to carbonate beers with two different pressures, you'll need a dual-body regulator. I have one (I make root beer @ 35psi).

Don't confuse this with a dual-gauge, which just means that there's a gauge for CO2 tank pressure and a gauge for beer tank regulated pressure.

You still want to get a manifold. Many uses for a manifold.
