Depends how cold the beer is and what volume of co2 you're shooting for.

Yeah that psi setting won't be affected by the co2 bottle/tank's temperature (mostly), but by the beer temperature. Lower psi levels needed the colder you get the beer.

You set your regulator to your wanted pressure whether it's located inside or outside your keezer. If I understand you correctly.
