Checking your carb levels

If I connect my beer line and draw a sample which comes out under carbed do I need to disconnect the line and drain? Or can I leave it attached and it will still carb? Also I have a hair over ten feet of beer line standard diameter; is it a waste to sample tough this or do people make smaller sample spigots?
 
Either way, there's beer in the line (not much - less than two ounces). You can disconnect it and drain and rinse it out, or leave it with the keg, hopefully cold, so the next time it'll be ready, and the first two ounces will likely be no more carbed than before.

I supposed some folks might "sample" with a picnic tap, but there's no big savings on those two ounces...
