With Dryhopping, carbonation comes off the beer. Is this carbonation significant when it comes to calculating priming sugar?
 
No. Any nucleation points will release co2, but not in any significant amount.

I never use priming sugar calculators as I've found them giving ridiculously high or low priming amounts at times, so if the amount seems "funny", not in the 3-5 ounce range by weight for 5 gallons, double check the input.
